BIOCHEMICAL AND BIOPHYSICAL RESEARCH COMMUNlCAJlONS GLUCOCORTICOID INHIBITION OF AMINO ACID TRANSPORT IN RAT HEPATOMA CELLS

چکیده
Received September 6, 1977 Summary: Dexamethasone rapidly and reversibly inhibits the initial rate of transport of a-aminoisobutyric acid in rat hepatoma cells in tissue culture. Colcemid and cytochalasin B neither inhibit transport nor interfere with its inhibition by dexamethasone, arguing that microtubules and microfilaments are not involved in this hormonal effect. Continuous protein synthesis is required both for the dexamethasone inhibition of transport and for its reversal, although cycloheximide alone inhibits transport in control cells by less than 25%. A model for the dexamethasone inhibition of amino acid transport is presented suggesting that glucocorticoids either block the synthesis or enhance the degradation of a rate-limiting protein in the transport system.

Loss of glucocorticoid regulation of plasminogen activator activity in anucleate rat hepatoma cells.
Glucocorticoids decrease the plasminogen activator activity of rat hepatoma cells through production of an inhibitor. We have examined the dexamethasone regulation of plasminogen activator in anucleate rat hepatoma cells to investigate the role of the nucleus in the steroid regulation of this membrane-associated phenomenon. Dexamethasone did not affect either the intraor extra-cellular plasmino...
متن کاملRegulation of insulin responsiveness in rat hepatoma cells.
Received November 28,1979 Summary: Insulin causes a 5 to lo-fold increase in the velocity of a-aminoisobutyric acid transport and a 2 to 3-fold increase in tyrosine aminotransferase activity in dexamethasone-treated hepatoma tissue culture cells. Maximal responses occur 2-4 hours after insulin addition but then decrease to control levels by 24 hours incubation. Medium conditioned by cells incub...
متن کاملHuman LAT1, a subunit of system L amino acid transporter: molecular cloning and transport function.
We report here on the cloning and functional characterization of human LAT1, a subunit of the amino acid transport system L. The hLAT1 cDNA, obtained from a human placental cDNA library, codes for a protein of 507 amino acids. When functionally expressed in mammalian cells together with the heavy chain of the rat 4F2 antigen (r4F2hc), hLAT1 induces the transport of neutral amino acids. When exp...
متن کاملHuman placental sodium-dependent vitamin C transporter (SVCT2): molecular cloning and transport function.
We report here on the cloning and functional characterization of human SVCT2, a sodium-dependent vitamin C (ascorbate) transporter. The hSVCT2 cDNA obtained from a human placental choriocarcinoma cell cDNA library, codes for a protein of 650 amino acids with a predicted molecular mass of 70 kDa. At the level of amino acid sequence, the human SVCT2 exhibits 95% identity to its rat homolog. When ...
متن کاملSelective targeting of nitric oxide synthase inhibitors to system y+ in activated macrophages.
Amino acid transport systems mediating uptake of nitric oxide (NO) synthase inhibitors were characterized in the murine macrophage cell line J774. Treatment of J774 cells with bacterial endotoxin (LPS, 1 microgram ml-1, 24 h) selectively increased the transport capacity for NG-monomethyl-L-[14C]arginine (L-NMMA), whereas transport of NG-nitro-L-[3H]arginine (L-NNA) was unaffected. Inhibition st...
